El lugar donde se encuentra el Palazzo Vecchio, era en el pasado un teatro romano. Descubrimientos arqueológicos a principios de este siglo revelaron estancias subterráneas que poseen restos de diferentes épocas.
Para finales del siglo XIII se decide construir un edificio que protegiera a los magistrados. La edificación se le encargó a Arnolfo di Cambio, quien empezó a construir en el año de 1299.
Para ese entonces el palacio se llamaba Palacio de los Priores.
Al morir Arnolfo en el año 1302, la construcción la finalizaron otros maestros de obra en 1314. Las arcadas que existían previamente del teatro romano se usaron como prisiones.
La apariencia del palacio actual es el resultado de muchas construcciones y ampliaciones que tuvieron lugar para los siglos XIII-XVI. En los años 1440-1460 se realizaron otras remodelaciones de envergadura ordenadas por Cosme «el Viejo», en donde la decoración del Renacimiento hizo acto de presencia.
Entre los años de 1540-1565 el palacio sirvió como residencia del duque Cosme I de Médici. Bajo sus órdenes se amplió el edificio, creó apartamentos que sirvieran de alojamiento para la corte, visitante y para la familia real. Cuando el duque se trasladó al Palacio Pitti como su nueva residencia, el palacio que anteriormente le sirvió de vivienda adoptó el nombre de Palazzo Vecchio (o Palacio Viejo).
